چکیده

Therapy-related acute myeloid leukemia (t-AML) is a heterogeneous group of myeloid neo‐ plasms occurring as an overwhelming complication in patients receiving previous cytotoxic chemotherapy and/or radiation therapy used to treat haematopoietic or solid malignancies or associated with immunosuppressive treatment for non-neoplastic rheumatologic/autoim‐ mune diseases or solid organ transplantation (Offman et al., 2004; Kwong, 2010). T-AML is an increasingly recognized condition included in the category of therapy-related myeloid neoplasms in the revised WHO classification of tumours of haematopoietic and lymphoid tissues, together with therapy-related myelodysplastic syndrome (t-MDS) and myelodys‐ plastic/myeloproliferative neoplasm, that constitute a unique clinical syndrome (Vardiman et al., 2009).

TET2 mutations in secondary acute myeloid leukemias: a French retrospective study.

Ten-eleven translocation 2 (TET2) mutations have been involved in myeloid malignancies. This retrospective study aims at evaluating the frequency and impact of TET2 mutations in 247 secondary acute myeloid leukemia cases referred to as myelodysplasia-related changes (n=201) or therapy-related (n=46) leukemias. Mutation of at least one copy of the TET2 gene was detected in 49 of 247 (19.8%) pati...

Retrovirus-mediated gene transfer of MLL-ELL transforms primary myeloid progenitors and causes acute myeloid leukemias in mice.

The MLL-ELL fusion gene results from the translocation t(11;19)(q23;p13.1) that is associated with de novo and therapy-related acute myeloid leukemia. To study its transforming properties, we retrovirally transduced primary murine hematopoietic progenitors and assessed their growth properties both in vitro and in vivo. MLL-ELL increased the proliferation of myeloid colony-forming cells in methy...

The incidence of secondary leukemias.

BACKGROUND AND OBJECTIVE The term secondary leukemia is usually employed to indicate both forms of acute myeloid leukemia (AML) evolving from previous myelodysplasia and forms of acute leukemia developing after exposure to environmental or therapeutic toxins or radiation (therapy related). Secondary leukemias account for 10-30% of all AML. The majority of secondary leukemias resulting from the ...

RUNX1 DNA-binding mutations and RUNX1-PRDM16 cryptic fusions in BCR-ABL+ leukemias are frequently associated with secondary trisomy 21 and may contribute to clonal evolution and imatinib resistance.

Acquired molecular abnormalities (mutations or chromosomal translocations) of the RUNX1 transcription factor gene are frequent in acute myeloblastic leukemias (AMLs) and in therapy-related myelodysplastic syndromes, but rarely in acute lymphoblastic leukemias (ALLs) and chronic myelogenous leukemias (CMLs). Among 18 BCR-ABL+ leukemias presenting acquired trisomy of chromosome 21, we report a hi...

Therapy-related myeloid neoplasms.

Session 5 of 2007 Workshop of the Society for Hematopathology/European Association for Haematopathology focused on therapy-related myeloid neoplasms. This report discusses the diversity and relevance of clinical, pathologic, and genetic features and provides an update on the pathogenesis of these disorders. We highlight common diagnostic issues such as the differentiation between therapy-relate...
